On the topic though, imagine a vampire girl who still believes in some obscure heresy or extinct religion like gnosticism or nestorianism. I only got exposed to the joke by hearing Jewish people making it. It makes more sense anyway as they tend to fixate on Jesus more when mocking Christianity due to him being the main difference. Shinobu does show some aspects of medieval mindset. However, it is typically for short gags like her saying stuff about young girls marrying. It doesn't amount to much more than that and she actually acts quite modern in many ways. Most lolipires are like this sadly, likely due to the author not wanting to make it an overbearing plot point. As you mentioned, humans changed a lot from the enlightenment so a proper vampire girl should be wildly different. It would be nice if someone researched medieval wedding ceremonies and made a story where a vampire girl is super picky about having one exactly this way. Maybe she could even take part in some traditions for holidays and other events and do things a little differently due to her age. You could probably find many funny things too to make her do. I think people for instance used to use their pee in some places to help clean clothing. A vampire girl would easily hold on to a lot of old ways and beliefs over the centuries due to the fact that the species doesn't really set itself up for regular social interactions.
