Neither are TUEEEEEEEEEEE, neither have a conveyor belt of suitors, demon lord is about the only part that it has in common with isekai, but then what? A sub genre by definition is subordinated to the main genre, this means the core defining elements are the same, like how punk has cyberpunk, biopunk, dieselpunk, post cyberpunk and so on. As long as the core principles of punk remain they can all be perceived as subgenres however "native isekai" would be in direct contradiction with a core defining genre trait. There is no subordination so no, it wouldn´t even be a subgenre of isekai, if anything it would be a variant under high fantasy but for that it would need, as mentioned, it´s own defining characteristics, structures and traits.

Native isekai is more aptly referred to "videogame/MMO inspired fantasy"
The plot structure and all that goes with isekai is not in the story but it uses the same aesthetics and tropes that a video isekai would. Adventure guilds, rigid classes, levels or somesort of advancement in the ranks, glowing skill activations with cooldowns, a maou and gaudy looking weapons and armor.
I don't personally think Frieren fits into these tropes enough to be classified as a mmo style fantasy anime but it has bits like how they show the dungeon map spell looking like a video game minimap or how the taverns/restaurants all serve standard Japanese style desserts and food.
